B3 - Relationship of leaky gut-derived bacterial translocation and genetic regulation of body fat distribution

The proposed study will test the hypothesis that bacterial load and composition in AT reflect increased intestinal permeability, leading to the inflammatory status and dysfunction of AT. Moreover, these events are linked to the metabolic sequelae of obesity and altered body fat distribution in interplay with the host’s genetic background.

Studies will evaluate mesenteric, visceral, epiploic and abdominal subcutaneous AT samples from 100 metabolically characterized obese individuals with and without evidence of AT inflammation and 25 lean controls for presence and type of bacterial DNA 16S rRNA gene sequencing. To verify viable bacteria, catalyzed reporter deposition - fluorescence in situ hybridization (CARD-FISH) and scanning electron microscopy will be used. A Lactulose mannitol test will be performed to measure intestinal permeability as a potential trigger of bacterial translocation, and biomarkers (e.g. calprotectin) indicating intestinal permeability will be assessed. To evaluate whether the link between genetics and metabolic phenotypes related to adverse fat distribution is mediated by AT bacteria, genotypes and expression of genes associated with abdominal obesity will be analysed concerning the individuals’ metabolic and inflammatory state and bacterial composition in AT. Finally, we will test the potential effect of bacterial presence in AT on adipocytes’ biology by treating human adipocytes with bacterial DNA and species affiliated in the bacterial taxa closely related to the studied subjects' inflammatory and adverse metabolic profile.

My PhD project is about epigenetics in metabolic diseases, especially the methylome alterations after weight loss interventions such as diet or metabolic surgery. I'm further establishing in vitro methods for the targeted SNP and DNA methylation editing using the CRISPR-Cas9 system.

Title: Tissue-specific cell-free DNA load in different stages of insulin resistance and diabetes remission in people with obesity
My Phd project is about whether we can use circulating cell-free DNA from liquid biopsies (blood) to capture tissue-specific DNA methylation structures and epigenetic (re-)programming, which will help distinguish between metabolically healthy and unhealthy states of obesity. Furthermore, I would like to identify differences in those cfDNA methylation patterns which characterize individual diabetes remission in response to metabolic surgery.

PhD project description:
I am interested in characterizing the inflammatory reaction of endothelial cells in response to molecular patterns associated with cardiogenic shock. The project focuses on the impact of CS-linked circulating biomarkers on multiple cellular components, and cellular systems. The integration of all four CLIP parameters is proposed to be conducted within hypoxic conditions. Moreover, a meticulous examination is undertaken to decipher the regulatory miRNAs targeted by IL-6 and other CLIP markers in the cohort of cardiogenic shock patients. The validation of these findings is intended through the application of bioinformatics tools.
