Z3 - Proteom- und Lipidom-Analysen mittel Massenspektrometrie und NMR

Das Z3-Projekt stellt eine state-of-the-art analytische Plattform für Proteomics und Metabolomics (insbesondere Lipidomics) zur Verfügung, die eine große Bandbreite moderner massenspektrometrischer und NMR-spektroskopischer Verfahren umfasst. Zur Charakterisierung der Proben von adipösen Probanden werden funktionelle Analysen des globalen Proteoms (Nachweis bestimmter Adipokine im Serum, globale Daten-unabhängige Serum-Proteomics), der Lipidzusammensetzung sowie des metabolischen Flusses durchgeführt. Zusätzlich werden wir räumlich aufgelöste Untersuchungen zur Metabolitverteilung mittels MALDI-Imaging durchführen.

(B) Mittels NMR können auch die Positionen der Veresterungen bestimmt werden. In (2A) ist der Einfluss unterschiedlicher Diäten auf die Fettsäurezusammensetzung von braunem Fettgewebe (Maus) dargestellt. In der verwendeten x:y Nomenklatur gibt "x" die Zahl der C-Atome in der Fettsäure an, während "y" der Anzahl der Doppelbindungen entspricht. Die Positionsbestimmung der einzelnen Fettsäurereste in den Triacylglycerolen ist mittels 13C-NMR-Sektroskopie möglich (2B). In vielen Fällen kann hier zwischen sn-1,3- und sn-2-Position differenziert werden.
